TC-2153 is a selective inhibitor ofstriatal-enriched protein tyrosinephosphatase(STEP), with psychotropic activity and low acute toxicity. TC-2153 increases the expression ofbrain-derived neurotropic factor(BDNF) in the brain. And it decreasesMAOAand5-HT1A receptors mRNA level in midbrain. TC-2153 also inhibits5-HT2A receptor-mediated signaling^[1].

(In Vitro)

TC-2153 (10 μM; 3 h) 减少皮层和海马神经元体外培养中DOI(HY-103124) 诱导的 c-fos 阳性神经元百分比^[1]。

体内研究
(In Vivo)

慢性 TC-2153 处理: TC-2153 (20 mg/kg; 腹腔注射; 每天 1 次，连续 7 天) 显示抗抑郁类效果，不改变运动，探索活动和焦虑情况^[1]。
急性 TC-2153 处理: TC-2153 (10 mg/kg和20 mg/kg; 单次剂量) 抑制小鼠体内 5-HT2A 受体的功能活性^[1]。

Animal Model:	SPF-state adult mouse males^[1]
Dosage:	10 mg/kg and 20 mg/kg; with or without 1 mg/kg DOI i.p. at 3 h after final treatment
Administration:	Oral gavage or intraperitoneal injection; single dose or once daily for 7 d
Result:	Showed antidepressant-like effect in the forced swim test without any adverse side effects on locomotor activity, anxiety, exploration, motor skill and obsessive-compulsive-like behavior. Both acute and chronic methods inhibited the functional activity of 5-HT2A receptors.
