VU0810464是有效的且选择性的非尿素蛋白门控的内向整流钾通道 (GIRK, Kir3)激活剂。VU0810464对神经元GIRK1/2 (EC50=165 nM)和GIRK1/4 (EC50=720 nM) 神经通道具有纳摩尔效价,并且具有脑部渗透性[1][2]。VU0810464(0、0.1、0.3、1、3、10、30μM)在SAN和HPC细胞中产生浓度依赖性的电流反应曲线,此外,VU0810464对神经元中Kir3通道激活的效力比SAN细胞高9倍。VU0810464对雄性C57BL/6J小鼠产生剂量依赖性的SIH响应降低(腹腔注射;30mg/kg,10mg/kg;30mg/kg;预处理30分钟)。
