用pan-caspase抑制剂IV和炎性caspases抑制剂Z-WEHD-FMK处理感染的细胞,几乎完全阻断了沙眼衣原体引起的golgin-84的裂解。通过使用Z-WEHD-FMK处理感染的细胞阻断golgin-84的裂解,导致高尔基碎片的缺失和一些感染性细菌2-log的减少。
用Z-WEHD-FMK处理感染细胞,有效地阻止了高尔基体裂解,用DMSO作为对照,然后用荧光神经酰胺标记细胞。共聚焦图像显示,在DMSO处理的细胞中,神经酰胺迅速进入包涵体膜,并且在细菌膜内包涵体中积累。相反,Z-WEHD-FMK处理的细胞只有轻微的荧光,因为大多数脂质在包涵体外的高尔基体样结构中积累[1]。
通常的caspase抑制剂(Z-Asp-CHz-DCB)和capase-5抑制剂(Z-WEHD-FMK)不能诱导ECyd处理的rRNA裂解。Caspase-5(ICErei Ill/TY),是ICE蛋白酶的成员,激活的通路可能与ECyd诱导rRNA裂解有关[2]。

Applications | Z-WEHD-FMK (80 μM; 9 h) elicited a near-complete blockage of C.trachomatis-induced cleavage of golgin-84 and increased GM130 expression in cells. Z-WEHD-FMK treatment resulted in a 2-log reduction in numbers of infectious bacteria, which could be reversed by knocking down golgin-84. Thus, Z-WEHD-FMK could effectively prevent Golgi fragmentation, and was used to explore the role of fragmentation of the Golgi compartment inChlamydiareproduction. |
