试验操作

Cell experiment:[1]

Cell lines

Human glioblastoma multiforme (GBM), rat prostate cancer (PC) and normal mouse 3T3 cells

Reaction Conditions

1 ~ 14 mM sodium ascorbate

Applications

Sodium ascorbate considerably decreased the proliferation and motility of GBM and PC cells. This effect was accompanied by intracellular ROS over-production and necrotic death of tumor cells, apparently resulting from their "autoschizis".

Animal experiment:[1]

Animal models

Male Wistar rats, 200 ~ 230 g

Dosage form

1 or 2 mg/kg

Injected intravenously at 3rd and 7th day after U87 cell implantation

Applications

In U87 tumor-bearing rats, intravenous administration of sodium ascorbate inhibited tumor invasionin vivoand seemed to reduce the size of neoplasia. Concomitantly, blood analyses did not show any signs of hemolysis or biochemical disorders after sodium ascorbate administration. High doses of sodium ascorbate were demonstrated to interfere with the expansion of GBM cellsin vitroandin vivo.
