Sulfo-NHS-Biotin (N-羟基磺酸基琥珀生物素)是一种胺反应性的生物素化试剂。最常见的胺反应性生物素化试剂可分为两种，N-羟基琥珀酰亚胺（NHS）酯和羧酸盐。通常，NHS酯-生物素化试剂包含反应性的NHS环结构，可以与蛋白或其它分子羰基上的胺反应，通过亲核攻击形成稳定的酰胺键，并释放NHS基团。由于NHS-biotin在水溶液中的不溶性，通过在NHS环结构上加入一个带负电荷的磺酸基，从而获得NHS-biotin的水溶性类似物sulfo-NHS-biotin。Sulfo-NHS-Biotin可以直接加入到反应性水溶液中，而不需要提前溶解在有机溶剂中。

o Irreversible—forms permanent amide bonds; spacer arm cannot be cleaved
o Very short—spacer arm is 13.5 angstroms; it consists of the native biotin valeric acid group only
