IL-4 is a pleiotropic cytokine produced by activated T lymphocytes, basophils, mast cells and bone marrow stromal cells. IL-4 plays an important role in the regulation of isotype switching, differentiation of precursor T helper cells and induction of IgE production in B lymphocytes. Also, IL-4 regulates cell proliferation, survival and gene expression. Excessive IL-4 production by Th2 cells is associated with elevated IgE production and allergy.
The murine IL-4 cDNA encodes a 140 aa protein with a 20 aa signal sequence. Mature mouse IL-4 shares 39%, 39%, and 59% aa sequence identity with bovine, human, and rat IL-4, respectively. Human, mouse, and rat IL-4 are species-specific. Recombinant murine IL-4 is a 13.5 kDa globular protein containing 120 amino acid residues.
