Interleukin-10 (IL-10) is an immunosuppressive cytokine produced by macrophages, monocytes, T cells, B cells and keratinocytes. IL-10 inhibits the synthesis of proinflammatory cytokines such as IL-1 and TNF-α and proliferation of T cells. IL-10 enhances humoral immune responses and attenuates cell-mediated immune reactions. It also enhances B cell survival, proliferation, and antibody production. IL-10 can block NF-kappa B activity and is involved in regulating the JAK-STAT signaling pathway. IL-10 plays an important role in the control of viral infections and allergic and autoimmune inflammation.
The human IL-10 cDNA encodes a 178 aa protein with a 18 aa signal sequence. Human IL-10 is active on murine cells, but murine IL-10 is inactive on human cells. Recombinant human IL-10 is an 18.6 kDa protein containing 161 amino acid residues.
