CFI-402257 HCl (CFI402257), the hydrochloride salt of CFI-402257, is a novel, highly potent, orally bioavailable and selective inhibitor of TTK protein kinase (Monopolar spindle 1 [Mps1]) with potential anticancer activity. It inhibits TTK with a Ki of 0.1 nM. TTK is an essential chromosomal regulator overexpressed in aneuploid tumors, and is important in regulating cell growth. High TTK levels correlate with a high tumor grade11 and poor patient outcomes. TTK inhibition are associated with a disabled mitotic checkpoint, resulting in chromosome segregation errors, aneuploidy, and cell death.
