MLT-748 (MLT748; MLT 748) is a novel, nanomolar, selective and allosteric inhibitor of MALT1 (Mucosa-associated lymphoid tissue lymphoma translocation protein 1). MALT1 paracaspase is central for lymphocyte antigen-dependent responses including NF-κB activation. As an inhibitor of malt1 peptide cleavage, MLT-748 acts by mimicking trp580 in malt1, binding to the same allosteric pocket as mepazine, displacing trp580, and locking the protease in an inactive conformation.
