Desethyl chloroquine is the main deethyl metabolite of chloroquine. Chloroquine diphosphate is an inhibitor of autophagy and toll-like receptors (TLRs). Deethylchloroquine has anti-plasma activity.

Chloroquine was injected intraperitoneally in wild-type and Huntington's disease mice. LC-MS/MS is used to compare the levels of chloroquine and its metabolites in blood, brain and muscle tissue. Within 4 to 24 hours after the last dose, the concentration of chloroquine is lower (5-15M) compared to blood or muscle, but it is more stable in brain tissue. Within 24 hours after injection, the level of deethylchloroquine, the active chloroquine metabolite in the muscle and blood, decreased, while the level of deethylchloroquine in the brain decreased and increased slightly within the same time frame.
